    White clover transformation using WCMV coat protein gene mediated by Agrobacterium tumefaciens

    • 摘要: 以白三叶Trifolium repens L.吸胀种子的子叶为转化体,用农杆菌介导将外源的白三叶花叶病毒外壳蛋白基因WCMV转入白三叶,经过筛选、分化和再生,得到了具有卡拉霉素抗性的转基因植株.对这些植株进行PCR、Southern 和Northern分析,结果表明,外源目的基因已经整合到白三叶基因组中并且得到了表达.

       

      Abstract: The white clover mosaic virus coat protein gene was successfully introduced into white clover, Trifolium repens L., plants through their cotyledons, mediated by Agrobacterium tumefaciens. Transgenic plants resistant to Kanamycin were obtained after selection, differentiation and regeneration. All of the transgenic plants were tested by PCR, Southern blot and Northern analysis, which indicated that white clover mosaic virus coat protein gene had integrated into the plant genome and got expressed.
