    水稻橙叶病植原体16S rDNA基因的序列分析

    Sequence Analysis of 16S Ribosomal DNA of Phytoplasma Associated with Rice Orange Leaf

    • 摘要: 利用植原体16S rDNA通用引物对水稻橙叶病发病植株的DNA进行了PCR扩增和基因克隆.序列测定表明,PCR扩增的片段全长为1 853 bp,包括1 527 bp的水稻橙叶病植原体(RYL)的16S rDNA基因全序列、234 bp的邻近间隔区的序列及部分(92 bp)23S rDNA基因序列.序列比较结果表明,RYL的16S rDNA全序列与其他植原体的相似性在88%~95%,其中与America aster yellows(AAY,GenBank登录号X68373)最高相似性为95%.利用最大简约法构建的16S rDNA系统演化树结果表明:RYL与AAY亲缘关系最近,同被聚类为翠菊黄化组(16Sr Ⅰ).

       

      Abstract: One set of the conserved primers was designed to amplify a 1 853 bp fragment which included 16S rDNA(1 527 bp),adjacent spacer sequence(234 bp) and partial sequence of 23S rDNA(92 bp) by PCR from total DNA sample extracted from diseased rice as the amplified template.Nucleotide sequencing of the amplified fragment indicated that rice orange leaf phytoplasma shared nucleotide similarity about 88% to 95% with other phytoplasmas from data of GenBank,and a maximum similarity at 95% with America aster yellow phy...
