番木瓜环斑病毒的提纯研究

STUDY ON PURIFICArION OF PAPAYA RINGSPOT VIRUS

  • 摘要: 详细报道了影响番木瓜环斑病毒(PRV)粗提纯效果的几个主要因素(繁殖寄主种类,有机澄清剂,分散剂,病毒浓缩前的去渣离心力,浓缩病毒的超离心力等)的最佳选择,繁殖寄主以角葫瓜最好。该研究综合上述最佳选择而拟定的一次差速离心和一次30%蔗糖硫酸连续密度梯度离心3h的简单程序较好地提供了PRV提纯的病毒有典型核蛋白紫外吸收曲线,A260/A280=1.68,提纯产量为2.56mg/100g西葫芦病叶,所
    Abstract: Every bes t cho ice of several main factors affecting the crude purification of p ap ayaringspot virus(PRV)(including kinds of propagation host,organic clarification agent,dis pers io n agent,centrifugal force fo removing sediment before coneentrating thevirus,supereentrifugal foree for concentrating the virus)as reported detailedly in thispaper.Jelly melon(Cu cum is metuliferus)was found to be a better propagation hostthan the Zucchini Squash(Cucurb itapepo).A simple proeedure of one cycle of diffe-rent speed contrifugation and one cycle of Cs_2SO_4 continuous density gradientsupereentrifugation with sugar cushion was designed.The virus preparation showed atypical ultraviolet absorption curve for nucleoproteins,A260/A280=1. 68,the yield ofvirus was 2.56 mg/100 g of infected squash leaves and the virus preparation couldbe suitable for the further research on the serology and molecular biology of PRV.
