大花紫薇新品种‘紫婵’雌性不育的显微结构及营养生理特征

    Microstructure and nutritional physiological characteristics of female sterility in a new Lagerstroemia speciosa cultivar ‘Zichan’

      目的 解析大花紫薇新品种‘紫婵’Lagerstroemia speciosa ‘Zichan’雌性不育机理,为紫薇属种质资源利用与创制提供理论基础。
      方法 运用组织解剖学观测‘紫婵’胚囊发育的形态结构,运用生理生化方法测定柱头可授性和花芽营养物质含量及酶活性等。
      结果 ‘紫婵’大孢子母细胞在形成八核胚囊后,胚囊发育停止,卵细胞和助细胞开始降解,胚囊形成空腔并最终萎缩;‘紫婵’花蕾中可溶性糖和淀粉积累量均低于同期可育大花紫薇,可溶性蛋白含量在胚囊成熟期高于大花紫薇;八核胚囊和胚囊降解时期,‘紫婵’超氧化物歧化酶(SOD)活性分别显著低于大花紫薇66.14%和40.20%,过氧化物酶(POD)活性分别显著高于大花紫薇139.20%和348.65% (P<0.05),过氧化氢酶(CAT)的活性呈下降趋势,与大花紫薇表现相反。
      结论 ‘紫婵’八核胚囊形成后,胚囊消解萎缩导致雌性不育。推测‘紫婵’花蕾整体营养物质积累量不足、抗氧化酶异常积累可能是导致其不育的内在因素。

       

      Abstract:
      Objective To elucidate the mechanism underlying female sterility in the new Lagerstroemia speciosa cultivar ‘Zichan’, and to provide a theoretical basis for the utilization and innovation of Lagerstroemia germplasm resources.
      Method Histological analysis was employed to observe the morphological structure of embryo sac development in L. speciosa ‘Zichan’, and physiological and biochemical approaches were used to assess stigma receptivity, nutrient content and enzyme activity in flower buds.
      Result In L. speciosa ‘Zichan’, the megaspore mother cells ceased embryo sac development after the formation of eight-nucleate embryo sac, accompanied by the degradation of egg cells and synergids. The embryo sac became vacuolated and eventually degenerated. In comparison with fertile L. speciosa, L. speciosa ‘Zichan’ flower buds accumulated lower levels of soluble sugars and starch at corresponding developmental stages, whereas the soluble protein content was higher during the mature embryo sac stage. At the eight-nucleate and embryo sac degradation stages, the superoxide dismutase (SOD) activity in L. speciosa ‘Zichan’ was significantly lower than that in L. speciosa by 66.14% and 40.20% respectively, the peroxidase (POD) activity was significantly higher by 139.20% and 348.65% respectively (P<0.05), and the catalase (CAT) activity showed a decreasing trend, opposite to that observed in L. speciosa.
      Conclusion Female sterility in L. speciosa ‘Zichan’ is attributed to the degeneration and collapse of the embryo sac following the eight-nucleate formation stage. It is speculated that insufficient nutrient accumulation and abnormal antioxidant enzyme levels in flower buds may represent intrinsic sterility factors.
